2Plate settings
ΔCt = mean Ct(target) − mean Ct(housekeeping). For each transcript, the control group's mean ΔCt is the calibrator. ΔΔCt = ΔCt(sample) − control mean ΔCt; fold change = 2−ΔΔCt.
3Map samples to treatment groups
Give biological replicates the same group name. NTC is excluded automatically. Group order and colors are used in plots.

The dotted horizontal line marks fold change = 1. Choose biological samples, individual qPCR wells, or mean only for point display. Bars and error bars follow the selected point level. In Combined view, technical-well mode pools technical wells from all included plates, while biological-sample mode uses one technical-replicate mean per sample per plate. Statistical tests remain based on biological samples (or plate-level group means in Combined statistics).
